This course provides a detailed introduction to the fundamental principles and hands-on techniques of microscopy. Participants will learn about the essential components and configurations of microscopes, including light paths, resolution, and magnification, with a focus on achieving optimal image quality.
- Key topics include an overview of brightfield microscopy and its components, the Köhler illumination setup and alignment process, and an introduction to phase contrast microscopy. Through practical, hands-on sessions, you will gain valuable experience in setting up and using brightfield and phase contrast microscopes effectively.
- By the end of the course, you will have a solid understanding of microscope configurations, be able to apply the Köhler illumination alignment process, and confidently use phase contrast techniques for enhanced image clarity in various biological and scientific applications.

- Köhler (Practical)
- Know your Lens: Aberration, Lens Design, Working Distance, Refractive Index Matching & Nomenclatures/Labels (Lecture)
- Microscope Maintenance & Cleaning (Lecture/Practical)
- Contrast Techniques: Phase Contrast & Dark Field, DIC & Polarisation, Epifluorescence basics (Lecture)
- Phase Contrast, DIC and Polarisation (Practical)
- Lenses, Open Optical Bench, NA & Swinging Apertometer (Practical)
- Flexible session; this may run subject to remaining time available and may include:
- Epifluorescence microscopy (Practical)
- Cameras, Image Capture, considerations for image analysis: (Pixels, Chip Type, Resolution, Signal to Noise, Dynamic Range & Scaling) (Lecture)
- Understand about microscope components, configurations and light paths
- Understand about resolution and magnification and how to use the correct setups and why
- Be able to set up a brightfield microscope and its components
- Be able to perform the Köhler illumination set-up alignment process
- Understand when and how to use the phase contrast technique and phase contrast components
- Practical skills in microscopy
